DeMaSk is a computational tool that predicts the functional impact of single amino acid substitutions in proteins using a linear model. It leverages interpretable, directional substitution matrices derived from deep mutational scanning data, alongside evolutionary conservation and variant frequency metrics from homologous sequences. The software is available for local installation via Python or as an online query interface.
